    PhD Position in Pharmacognosy

    At the Department of Medicinal Chemistry, Division of Pharmacognosy. Starting date: February 2010, or as agreed.
    The open position is placed within the Cyclotide group at the Division of Pharmacognosy. The overall goal of our research is to explore the possibilities to capitalize on the extraordinary stable scaffold of the circular plant proteins called cyclotides, for engineering and production of pharmaceutical and agricultural agents. In particular potential medical applications of cyclotides, i.e. as anti-cancer and antimicrobial agents, and cyclotide biosynthesis are addressed. Specifically, today we focus our efforts to determine the structural plasticity of the cyclotide scaffold, by studies of natural occurrence and native structures; determine the biosynthetic pathway of cyclotides, by studies of the plant genome, the processing of the cyclotide precursor and cyclotide oxidative folding; and to establish cyclotide structure activity relationships, by assays of native cyclotides and synthetic mutants.
    Our group offers of stimulating and productive environment, with good support financially and a well established scientific network nationally and internationally.
    Suitable candidates may have a degree in Pharmacy, Science, Biotechnology or similar, with good knowledge in chemistry and biology. The project is multidisciplinary, and involves methods from the fields of biochemistry, molecular biology and analytical chemistry (e.g. peptide synthesis and purification, HPLC, MS, NMR, bioassays, genetic analysis and protein expression). Previous experiences of such methods and techniques may be of advantage. The applicant should have a strong interest for research and research methodology, have good communication skills, and be able to work in a team.
    The holderof a PhD-student position shall primarily devote her/his time to research studies (equivalent to 4 years full time research studies). Other departmental work, such as educational or administrative services can be part of the position (max 20 %). The applicant must be eligible for PhD studies. Priority is given to those who are deemed to show the greatest aptitude for a successful completion of graduate studies. The rules governing this process can be found in Higher Education Ordinance (Högskoleförordningen) 5 kap §§ 1-7.
    Local guidelines at Uppsala University determine the salary.

    PhD Scholarship in Genetic assessment of antibiotic resistance in animal herds


    At the National Veterinary Institute a 3-year PhD position in bacteriology is available from April 1st 2010.

    The candidate will be placed at National Veterinary Institute, to work in the Research Group for Bacteriology and Pathology, and will also work at the Department for Veterinary Disease Biology at KU-LIFE. A residency at a research institute in USA is planned as part of the project.

    Project description
    The aim of this PhD project is to explore and develop effective methods for assessment of antibiotic resistance in animal herds by molecular methods. The PhD-student will develop quantitative PCR test on genetic markers for antibiotic resistance and compare these methods with traditional methods for determination of antibiotic resistance by cultivation on selective media, investigations on indicator bacteria and assessment of the global effect on the intestinal microflora. The methods developed will be evaluated in model studies as well as in swine herds. The project aims at generating a method that can be used for a quick and effective documentation of herd resistance by certifying the low level or absence of certain antibiotic genes and for further recommendations regarding the use of antibiotics for production animals.

    Qualifications



    Applicants with the following qualifications are preferred:
    • A scientific degree in either veterinary medicine, biology or a comparable discipline.
    • Experience in bacteriology and molecular methods.
    • An interest in animal husbandry and antibiotic resistance.
    • Fluency in English
    • Good social skills, innovative and interested in communicating research results
    Approval and Enrolment



    The scholarships for the PhD degree are subject to academic approval, and the candidates will be enrolled in the Life Science programme of DTU. For information about the general requirements for enrolment and the general planning of the scholarship studies, please see the DTU PhD Guide.

    PhD position in Enzyme Development

    In the Marie Curie Initial Training Networks, LeanGreenFood, Dept. of Chemical & Biochemical Engineering, Technical University of Denmark seeks a PhD student to generate novel enzymes for production of oligosaccharides and hydrocolloids”. The aim of the LeanGreenFood project is to reduce the chemical treatment in the production of the food and food ingredients by utilizing enzymatic catalysis in the process. The specific objectives of the present PhD study are to discover new efficient enzymes capable of hydrolyzing specific bonds in plant cell wall polymers. Potential enzymes will be identified from fungal strain collections. A number of new genes will be identified and genetic constructs will be prepared and transformed into Bacillus and Pichia strains. Specific enzymes will be optimized by molecular evolution to increase the potential use in the bioreactors.

    Skills:

    • Candidates should have a master's degree in molecular biology, chemical engineering, biotechnology, biochemistry or a similar degree with an academic level equivalent to the master's degree in molecular biology.
    • A strong commitment for research
    The PhD student should have a strong interest in gene expression in bacteria or yeast. The work requires basic knowledge on genetic engineering and bioinformatics and PhD students with these experiences will be preferred. The position offers an opportunity to work in a dynamic multi-disciplinary research group with strong focus on the production of hydrolytic enzymes to generate hydrocolloids, fibers and prebiotic oligosaccharides from agricultural side-streams. Hence, there is ample opportunity to interact with colleges that specialize in fields like molecular biology, biochemistry, modelling, enzyme kinetics, fermentation and down-steam processing. Together this ensures a highly stimulating working environment with all the right competences to serve as a solid platform for this project.

    - Development and application of Fast multidimensional NMR methods for the study of molecular kinetics and excited-state conformations.

    - Studies of intact Bacterial Cell walls using solid- and-liquid state NMR.

Applicants will also have access to a fully equipped wetlab for isotopically labeled NMR sample preparation.

The Faculty is home to 3500 students and nearly 2000 staff and is internationally renowned for the quality of its research.

    The NMR Research Group is part of the Chemistry Department (http://www.chem.uu.nl) and belongs to the Bijvoet Center for Biomolecular Research (http://www.bijvoet-center.nl). A major research line (headed by Prof.Dr. Marc Baldus) concerns the development and application of solid-state NMR (ssNMR) methods in a biomolecular context. In contrast to other biophysical methods, ssNMR techniques can be used to study atomic structure even if molecules are not soluble or crystalline. Correspondingly, ssNMR offers new possibilities to study complex biomolecules such as protein assemblies and Amyloid proteins in a biologically relevant context. We are working in these fields of research and collaborate with internationally leading groups at Utrecht University and abroad.     Job description

    You will be part of a team working on the study of elementary aspects of protein folding and dynamic assembly by ssNMR. In particular, you will follow the structural and dynamical changes associated with Protein aggregation via polyglutamine stretches. This process occurs in a number of severe diseases such as Huntington's disease and represents a role model for other neurodegenerative illnesses such as Alzheimer's or Parkinson's disease. In parallel, you will study other Amyloid proteins and the process of biomolecular folding in a membrane environment in a comprehensive manner. These experiments will allow you to understand that atomic details of molecular folding and assembly that are of vital relevance for biomolecular function and its failure. The position is funded by a research grant from the Netherlands Organization for Scientific Research (NWO) to Prof. Dr. M. Baldus.

    Qualifications

    The successful candidate should have a Master's or equivalent degree in chemistry, physics, or biology with a general interest in biophysics and spectroscopy. Experience in NMR or a related spectroscopic method is desirable.

    Terms of employment

    We offer a position for four years. You are expected to have a Ph.D. thesis written by the end of the fourth year. The salary starts at 1956 and amounts to 2502 euro gross per month in the fourth year of the appointment. The salary is supplemented with a holiday allowance of 8% and a year-end bonus of 3% per year. We also offer a pension scheme. Employment conditions are according to the Collective Labour Agreement of Dutch Universities terms of employment. Next to research (90%) you are also expected to contribute to teaching (10%).

    Increasing evidence suggests that protein structure and function is strongly influenced by the surrounding molecular, eventually cellular environment. Solid-state NMR is an ideal method to investigate such heterogeneous samples and has made significant progress in determining three-dimensional protein structures. Yet, additional efforts are needed to streamline the 3D structure determination process and to relate data to the surrounding molecules. As a postdoc, you will be working in the ssNMR team to develop high-resolution methods to infer structural information in complex systems such as membrane proteins or cellular preparations. For this purpose, you will combine current ssNMR structure determination concepts with advanced labeling and computational biology techniques. Initial studies on all relevant aspects have already been published by the Baldus group. These experiments will create a novel means to study elementary biomolecular processes in high spatial and temporal resolution with direct implications for understanding cellular functioning and disease. The position is funded by a research grant from the Netherlands Organization for Scientific Research (NWO) to Prof. Dr. M. Baldus.

    Qualifications

    The successful candidate should have a Ph.D. degree in chemistry, physics, or biology with a general interest in biophysics and spectroscopy. Experience in NMR, in particular solid-state NMR, is desirable.

    Terms of employment

    The position on this project is available for a maximum of three years. Salary is dependent on qualifications and experience and ranges from ?2802.- to ?3678.- gross per month (salary scale 10). The salary is supplemented with a holiday bonus of 8% per year and a year-end bonus of 6.4% per year. We also offer a pension scheme. Employment conditions are according to the Collective Labour Agreement of Dutch universities, terms of employment. Next to research (90%) you are also expected to contribute to teaching (10%).

    Description of the Faculty / Research group

    The Faculty of Science consists of six departments: Biology, Pharmaceutical Sciences, Information and Computing Sciences, Physics and Astronomy, Mathematics and Chemistry. The Faculty is home to 3500 students and nearly 2000 staff and is internationally renowned for the quality of its research.

    The NMR Research Group, headed by Prof. R. Boelens, is part of the Chemistry Department and belongs to the Bijvoet Center for Biomolecular Research. The research within the NMR group is focused on the structure determination of proteins and protein complexes by high-resolution NMR spectroscopy and bio-computational modelling. The group has strong research lines on protein-DNA and protein-protein complexes in transcription and DNA repair and modelling of bio-molecular complexes. The NMR Research group also belongs to the European NMR Research Infrastructures, providing experimental NMR access to European groups.

    We are searching for a post-doc in biomolecular NMR.

    Job description

    You will be part of an international team using advanced post-genomic techniques for the discovery and development of new therapeutically relevant molecules issued from the venomous marine cone snail species Conus consors, a project funded under the 6th framework programme of the European Union (Contract no. LSHB-CT-2007-037591 – CONCO) (see also http://www.conco.eu/ ). Your role in this project will be to apply both solution and solid-state NMR methods for the characterization of small snail toxins and their interaction with membrane receptors.

    Qualifications

    You should have a Ph.D. degree in chemistry, physics, bioinformatics, structural biology or a related field. You should possess a strong research background in biomolecular NMR (solution and/or solide-state) with experience in the collection and interpretation of NMR data, and are expected to show initiative and be able to work independently.
